Potensi Senyawa Baikalein sebagai Inhibitor Tumor Necrosis Factor Alpha Converting Enzyme (Tace) melalui Molecular Docking
Abstract
Pendahuluan: Tumor Necrosis Factor-α (TNF-α) berperan dalam beberapa penyakit seperti peyakit jantung, dan metabolik termasuk gagal jantung, hipertensi, arterosklerosis, stroke, infeksi paru, diabetes melitus dan pelepasan sitokin tidak terkontrol pada infeksi SARS-CoV-2. Tumor Necrosis Factor-α Coverting Enzyme (TACE) berperan dalam pemotongan tmTNF-α dan melepas sTNF-α yang memberi sinyal melalui TNFR1 dan TNFR2 sehingga penghambatan TACE berpotensi sebagai pendekatan terapi beberapa penyakit tersebut. Tujuan: Untuk mengetahui potensi baikalein dalam penghambatan TACE melalui pendekatan in silico molecular docking. Metode: Penelitian ini meliputi beberapa tahapan yaitu preparasi protein, optimasi struktur 3 dimensi baikalein, validasi molecular docking dan docking baikalein. Hasil: Metode docking dinyatakan valid dengan nilai RMSD 0,61. Nilai energi ikatan hasil docking baikalein (-8,46 kkal/mol) lebih negatif dibandingkan native ligand Q283 (-7,86 kkal/mol). baikalein berinteraksi dengan TACE melalui ikatan hidrogen pada residu asam amino yang sama dengan native ligand Q283 (LEU348 dan GLY349). Kesimpulan: Baikalein berinteraksi lebih kuat pada situs ikatan yang sama dengan native ligand sehingga secara molecular docking senyawa baikalein berpotensi dalam menghambat TACE.
